Abstract
Hexavalent chromium (CrVI) is a toxic anthropogenically industrial material. This study developed and optimized a bacterial based biosensor for the soluble CrVI. The Bacillus megaterium producing chromium reductase was immobilized onto screen printed carbon electrode (SPCE) to electrochemically detect the CrVI in the solution. The experimental analysis indicated that bacterial density, electrolyte solution pH, and scan rate significantly affected the biosensor current (p < 0.05). Optimization revealed that the biosensor works best at bacterial density of log phase, pH 3, and a scan rate of 100 mVs-1 with a desirability of 0.923602, resulting in a cathodic peak current of -61.16 μ-A.
