Abstract
Acacia honey (Accacia crassicarpa) contains various bioactive compounds, such as phenolics and flavonoids that provide pharmacological effects, including anticancer activity. This study aimed to determine the potential of acacia honey from Indonesia in inhibiting proliferation and inducing apoptosis in WiDr colon cancer cells in vitro. Cells were exposed to various concentrations of honey and analyzed using the MTT method to measure WiDr cell antiproliferation, Hoechst staining for apoptotic morphology, and qPCR for apoptotic gene expression against BAX, BCL- 2, CASPASE-3, CASPASE-8, and P53 genes. The results showed that acacia honey at a concentration of 5.5% could inhibit WiDr cell growth by 64.34%. The 5% acacia honey can increase the BAX/BCL-2 ratio, and reach the apoptosis execution stage by increasing CASPASE-3 expression. In addition, acacia honey induced apoptosis through extrinsic and intrinsic pathways at concentrations of 2.5% and 1.25%, as indicated by increased expression of CASPASE-8 and P53. The conclusion of this study is that acacia honey inhibits cell proliferation, triggers morphological changes, and increases apoptotic gene expression.
